I don't think this is related to rhythmbox.
I just got this myself, but several programs (firefox, rhythmbox, movie player) 
will not play sounds.
I started a guest session (Ubuntu Karmic, 9.10), and sounds played, but when I 
switched back to my normal login, same error.
I think I was trying to run mythtv and it may have exited improperly and hosed 
my alsa port or something??  I'm not ready to reboot.

How odd.. I fired up Mythtv again, and it played sound....   I closed
it, and wow!  Now Rhythmbox & Firefox play sounds, too.

Sorry, my only thought for a fix is to try restarting the last program
that was playing sounds successfully, maybe it had a lock on something.
