Happened to me on Ubuntu 9.10 64 bit. It was fixed by:

1. Going to Ubuntu One online and remove any unneccesary overlaps- things with 
the same name. Keep your current account with Ubuntu One sync to desktop
2. After deleting, log off and log on again.
3. Then, make sure you are logged into ubuntu one online website and launchpad 
(I think logging into launchpad is unnecessary but it does not do any harm)
4. Go to Tomboy and go to preferences and  synchronization and change it to 
tomboy web. Click the Connect button
5. The webpage should load. Give the computer a different name than what has 
been assigned before.
6. If that doesn't work, redo these one more time and this time try to delete 
~/.config/ubuntuone/syncdaemon.conf if it exists. 
7. You should be connected but you might not be able to sync your notes. Go to 
https://one.ubuntu.com/notes/
8. If it shows a 500 error, wait for 5 minutes and reload the page.
9. Go to Tomboy and try Tools-> Sync Notes
10. It should work.

Hopefully there is a fix for all that. This bug was also overlapped by another 
one: https://bugs.launchpad.net/ubuntu/+source/ubuntuone-client/+bug/501308
The suggestions there didn't impact much since this is related to Tomboy.
