Thanks a lot for your help. I reinstalled U1 followinng the FAQ and both my computers synced again. The problem persists though and I think I might know what the problem is.
Both clients sync fine indepentendly. At home I use both computers simultaneously. As soon as I use them both at the same time, only the client works on which the service was started first. The other one shows that U1 is still running, but it won't even connect, let alone sync anything in any direction.
