I was initially having the same issue with an Epson Stylus c48 (but without the Unknown IEEE 1284.4 error number 66) when running inkblot as an ordinary user, although inkblot did work if i run it as root (sudo inkblot).
The website for Libinklevel (background code for ink and inkblot) mentions: "Users of programs which make use of libinklevel need read/write permission on the device the printer is connected to. For parallel port printers this is /dev/parport[portnumber] and /dev/lp[portnumber], for usb printers this is /dev/usb/lp[portnumber] or /dev/usblp[portnumber]."
