Hi Dan, I had the same problem but got it sorted by doing this: install esound in synaptic then move your sound file you want Thunderbird to play in to your home folder (not in any folder just direct in to home folder) then start Thunderbird and go to preferences and browse for your sound file in your home folder. That worked for me.
