I didn't get a reply from Luke but here as the basic things to test:
- boot the liveCD, enable screen reader, check that it's working in GTK3 applications (ubiquity, rhythmbox, gedit, shotwell, ...)
- install, reboot, log into the session, check those are still working
- watch for any performance or stability bug that lead to errors in a11y components

I don't have really specific items and I'm still trying to get details from luck but the key points are to check out the fact that accessibility is indeed working and to watch for issues that might be created by it being one (like errors.ubuntu.com issues pointed to at-spi or atk components)
