Any errors reported in, for example, /var/log/syslog or /var/log/samba/...? My first thought was a timing issue, i.e. the cifs kernel module isn't loading quickly enough. I'm not sure if this is the right avenue at all but one I would try and check. Have you changed anything (updated s/w) on either system recently? For example I recently upgraded my home server to 10.04 and a change to the samba stack stopped my Wife's WindowsXP VM (just for iTunes) working, but didn't affect anything else. It only required a minor change (forcing Samba to be a WINS server) but just to illustrate a point.
