Without more details (which architecture/board, how Linux/uClinux boots on it), I would probably guess you would need to generate a new ROMfs with whatever you need. GTK+ is a biggie, though, with lots of dependencies (which would add a lot to your ROMfs size, which can cause additional issues depending on your arch), so if you are new to uClinux, I would suggest trying something simpler first.
