If you're talking about using a GPU with guacd on the server running Guacamole, this is not currently supported - there is no direct support in guacd to take advantage of GPUs attached to the system. On the Windows Server side, you're probably looking for the GFX Pipeline (also known as RemoteFX) extension to RDP, which is in the process of being implemented.
