Hi there, I have developed a mail clustering solution that works with most mail servers, and has massive scalability. It's called Proto Balance Mail and it's downloadable from http://protobalance.com/protobalancemail.html
Now Proto Balance Mail directs traffic, and aportions mailboxes -- so it does not actually store mail. It is meant to allow you to deploy multiple instances of whatever mail server and webmail client you are already accustomed to. It would be good to integrate this product with RoundCube of course. The result would be a web-mail solution that supports large sites - like those with over 10,000 or 100,000 mailboxes or more.
