Just an extra tip for anyone experiencing trouble with TileCache/mod_python:
We had a similar problem with TileCache, and couldn't get mod_python to work on our Windows server. We switched to geoCache [1], which basically is a port of TileCache from Python to C#, and this works very well for us and our Windows IIS-server. It integrates very well with OpenLayers.
