If you mean open an Excel spreadsheet and enter more data to it and save it again in Excel format, yes, you can do that with OpenOffice.org Calc. If you do, you will probably get an informational message when you save, warning you that saving in formats other than the native Open Document Format (ODF) may result in some formatting losses or changes. This is true but, in general, should not affect basic Excel spreadsheets, so you should answer "No" to the prompt to save in ODF.
