Of course it will. 

spamc will send the message to your daemonised copy of spamd.
Or you can switch to the same used as spamd runs under and run 

spamassassin -usual_commandline_parameters -D debug options
<message_to_be_tested

I've done just this for years with a copy of SA installed on a
development machine which is set up to duplicate my live SA
installation. How else can I develop rules and test changes without
impacting live operation? Needless to say, I also have an automated way
to transfer configuration files etc to the live site and restart spamd.

Its all documented as you should know if you've read the manpages and
the manual on the SA website.
