According to http://www.varnish-cache.org/docs/2.1/faq/general.html you can't.

But always you can do a GET and look at X-Varnish header, hit (two
numbers) or miss (one number), says to you if the object is in cache,
of course only if the object is cacheable.

And of course, if the object is cacheable and the object was not in
cache before you do GET, now it's in cache :) In some scenarios this
trick is not desirable, specially if you ONLY want to know if the
object is in cache.
