You can mount the usb key as ISO-cd and then boot from that ISO as CD-ROM.
But I suppose that is not what you really want?

Not really - the problem then is that (I expect) VirtualBox will treat
it as read-only.

What you may done then is :
   1.    install the host system to boot from the usb
   2.   boot the host from the usb key
   3.   install VB on the usb key
   4.   install the guest on the usb key
