Jimi,
I'm fortunate to have these skinny little fingers and I can just pick up a
32 and stick it in the vise.  For people with big fingers, I suggest a
locking forceps or a good set of tweezers.  A set of reverse tweezers might
work for you.  They open when you squeeze them, and stay shut normally.

As far as for being one-handed, putting the hook in your vise is only the
1st hurdle to be jumped.  Have fun.  I can tie flies without a vise, but not
one-handed.
