Surely that starts with "let", as in

     let NERDTreeBookmarksFile='/home/myuser/.vim_nerdtree_bookmarks'

I know little about NERDTree, but it looks like you want

    let NERDTreeBookmarksFile = $HOME . '/.vim_nerdtree_bookmarks'
