Yes, by setting up filetype associations which use Vim's --remote series of 
commands. See :help --remote and following.

http://vim.wikia.com/wiki/Launch_files_in_new_tabs_under_Windows has it set up 
for multiple tabs within one Vim instance. It is easily adaptable for multiple 
buffers in a single window instead.

The wiki article relies on --remote-tab-silent to launch files. If you don't 
want tabs, just use --remote-silent.
