You ask a good question but the explanation can be somewhat voluminous, to say 
the least.  

In short, replying to a tweet means that anyone who views the tweet can, should 
they choose, read your reply.  Also, your reply will be added to your profile, 
in the tweets and replies area.

Should you choose to quote a tweet, however, doing so is the same as 
re-tweeting but with your added comment placed before the original tweet.  This 
means that your quoted tweet with comment will be placed in your 
timeline/profile and anyone following you will see the quoted tweet with 
comment.

As for the period technique, yes, it does work and I used to use this technique 
frequently before gaining a better understanding of the TwitterVerse.  IN 
short, as long as the @ symbol does not appear at the beginning of your tweet, 
everyone can see it.  Conversely, if you place the @symbol at the beginning of 
your tweet, then only followers of that particular profile will see your tweet. 
 

Generally, I recommend that you do not place any @ symbols at the beginning of 
any tweets.

It takes time to understand Twitter so be patient with yourself and enjoy.

What are the differences between these two options? For those of you that tweet 
frequently, how do you use reply or quote tweet? 

Also, in researching this topic, I came across a tip where you can put a period 
before the at sign of an user, so that reply will be seen by everyone. Has 
anyone done this?
