I haven't listened to an audio book through Libby yet, but I did check out and read a Kindle book in the Kindle app which I checked out with Libby.
You don't need to wait for a physical book to become available, but it works the same way. Only a certain number of books are allowed to be checked out at any one time. If a book is on hold then you need to wait until a book is returned before you can check it out. It's more like a counter that gets incremented and decremented whenever a book is checked in or checked out.
