Probably the book is marked up incorrectly or there is some other reason iOS thinks it is in another language. Try going into settings, general, accessibility, VoiceOver, language rotor. On this page, find U.S. English (or your preferred variety of English) and if it isn't selected, double-tap it to select it. This makes an additional option available in the VoiceOver rotor called language. The rotor will contain any languages you have selected in the settings as well as a "default language" option, which detects the language automatically.
Now go back into iBooks and select Language in the rotor. Change it from default language to English. I bet that'll fix it.
