Yes, as I said, your home screen layout is backup and restored and that includes where the apps are and any folders which exist.

WiFi sync does require iTunes and it does not only require the computer to be on, but also for iTunes to be open. It really is no different than plugging your phone into the computer and syncing with iTunes except that the sync is done via WiFi. The computer with iTunes and the phone also have to be on the same WiFi network. I personally always found it somewhat slow and a a bit unreliable, sometimes it would work and other times it would not. I actually haven't tried it in quite a while so maybe it's more stabil now, but to me it's just as easy to plug the phone in. One thing I am not sure of is whether Windows XP even supports the very latest version of iTunes and WiFi sync. I assume it may, but since official support for Windows XP stopped over 2 months ago it really is a good time to consider upgrading. Microsoft does no longer release critical security patches for XP and I would not want to take the chance. If the phone is sinked via wifi, does iTunes have to be kept on the computer? 5. does the computer have to be on while the phone is being sinked wirelessly?
