Hi Steve, I'd personally triple-click home on the 6 Plus when it's at the set-up screen and do the initial set-up via VoiceOver on the device. But if you want to do it via the Mac, press the button in iTunes labeled iPhone and you should find the new 6 Plus there, complete with a web-page type screen you interact with to take you through it.
