When James Best auditioned for the role of Jim Lindsey, he was asked whether or not he could play the guitar. His honest response was "I own TWO guitars." Later, when it came time to shoot the episode, Earle Hagen (the music director) handed him some sheet music and said "Here's the music you'll be playing for this episode." At that point, James had to confess that he wasn't a "real" guitarist and that someone else would have to do the actual playing. So the answer to the question is yes, James Best really could play the guitar (at least a little bit) but he did not do his own playing on The Show.
