I'd go two column, personally. I think the content area is too wide. Anything that requires the eye to track past about 500px or it's equivalent becomes increasingly more difficult to read. You could do a fixed width layout in em's to still provide for accessibility, while giving you a little more control over the layout too.
I'm not so enamored with drop-downs, though sometimes they're a necessary evil (more often than that though, they're an indicator of poor IA).
