Is it happening locally? Did you install MySQLdb?
I app.yaml you have to add under "libraries"
- name: MySQLdb
version: "latest"
Finally you have to add your MySQL credential to dev_appserver.py like:
dev_appserver.py [root folder of web2py] --mysql_user=[mysql user]
--mysql_password=[mysql password]
