What Chuck said.  

But to elaborate: 1) You can check the format.  2) You can check the domain for 
an MX record. (Be aware step 2 can take several seconds or more.) and 3) send 
an email.  Anything other than that is pointless.

You could try to check the address at the mail server with VRFY. That is what 
the command is intended to do, but no sane mail server admin enables it. 
Spammers would build lists by guessing randomly.  

If VRFY is off, then using RCPT TO without sending an email is considered 
abusive and will generally get you blocked by the mail server and blacklisted.  
Not only that, but many mail servers are configured to receive mail for any 
address, whether it exists or not, so even RCPT TO is pointless.

So... send an email and make the recipient click something before giving them a 
cigar.  It's the only way to be sure. (^_^)
