Here is one way:

urls = (
    '/mailing(?:/(.*))?', 'newsletter'
)

But your approach is more readable than this.

It can be simpler if you are willing to take '/' as part of the action.

urls = (
    '/mailing(/.*)?', 'newsletter'
)
