As mentioned, Ricky Gervais is very funny, but is no
longer free. I think it's $2 a week now (they WERE
free, but their deal with The Guardian was only for 12
episodes). But Ricky and Stephen Merchant mocking Karl
Pilkington and his round, round head and daft diary
are good laughs.

NPR's Wait Wait Don't Tell me is available via
podcast. I always miss it when it's broadcast, so
podcasting has saved my life.

Also Jack Magnan's Deadpan is interesting. It's not
100% humor, he does a lot of interviews (he did one
with Mike Nelson from MST3K that was really good)

I heard good things about Jack Black's podcast Nacho
Libre, but never downloaded it.

Also, iTunes has a comedy area in the podcast, so you
could try a few and see what appeals to you.

Good luck, and if you find other good ones, be sure to
tell the rest of us!
