If you are using the 'NT Service' plugin, it uses the Win32 API to check an
NT Service. This API uses NETBIOS (probably NetBIOS over TCP) to see if
the service is running. The up/down status returned by this check is
whether or not the service is running -- just like you would see in the
'Services' section of the control panel. What this could mean is that a
service is 'running' and therefore considered up, even if it is
malfunctioning and not answering requests from other systems on the
network.
This is in contrast to the 'TCP/IP Service' plugin, which actually connects
to a application port on the remote system and makes a request of it --
expecting a valid response.

Does anybody knows how WUG is doing the actual checking of the nt service?
Does it use that particular port or something else?
Thanks,
