Yes, you can.

Open the mail action.  
Click the 'Options' tab at the top of the screen.
In the "Custom header lines:" section add your "Reply-To:"

I have this code in the "Custom header lines" section (in T2K, we're
upgrading to 5.5)

<@IFEMPTY @@local$student_email>
<@ELSE>
Reply-To: @@local$student_email
</@IF>
