Wander through the bugs listed there that have a resolution of "fixed" and verify that they are indeed fixed. You can also look for collateral damage in the areas affected by the fixes.

Both of the default plugins saw some change and could use extra testing.

Areas that often hurt us are xmlrpc and pingbacks. Give those some exercise.

If you want to get fancy, turn off referrers using the web developer extension in firefox (or whatever you tool you prefer), and verify that redirects still work. Update options and create/edit/delete posts and cats and make sure you are redirected properly after each operation.

Free form testing is also good. Start clicking around and see what falls off. Make sure your usual workflow still works.
