I just tested 2.6b3 on a fresh site install and it worked fine for me. I created a new (empty) DB, DB user, and DB password, put them in the wp-settings.php file, and it worked fine.

This was on my Ubuntu Linux test box, running Apache 2.0, PHP 5.2, and MySQL 5.0.

Are you sure you (a) have actually created a DB and DB user, and (b) have entered the correct host for the DB, as well as the right DB information? I guess if you have set up WP quite a few times, you probably know what you are doing, but anyone can occasionally make mistakes. But I don't think WP 2.6b3 is the problem...
