/opt/local is typically MacPorts, not brew (brew installs mostly in /usr/local, 
and MacPorts prefers that possibly conflicting programs NOT be installed in 
/usr/local).

I have XQuartz 2.8.1 from https://www.xquartz.org on currently updated (not 
beta) Intel Monterey (the only Monterey I have is a VM under Parallels), and it 
works fine, whether I start it first by clicking on the XQuartz icon, or 
whether I start it on demand by running an X11 program from Terminal (or iTerm2 
or whatever).

One can install X11.app from MacPorts (ends up in 
/Applications/MacPorts/X11.app) as well. I don’t have that on Monterey, but I 
have both on Catalina, and one can even run both at once (they have different 
DISPLAY variable values, only one of which can be preset depending on what 
LaunchDaemon startups are enabled I guess). Sometimes it’s claimed that 
MacPorts is more current; but depending on when I’ve checked, that’s not really 
been an issue, and the “simplest” case for comparison with most people is 
probably using what’s on www.xquartz.org.

Maybe you need to uninstall/reinstall XQuartz; or maybe it’s something else. 
But it CAN work.
