Yes, you can, to get more convenient interface you can use Asterisk::AGI.

You need primarily Asterisk::AGI - AFAIK it is not Debian packaged.
It is a separate glue (so-called "binding" but in fact only trivia stdio glue
in the case of Asterisk) between Asterisk itself and Perl:
