No, that's all one step - prepare and test a package, and then file this bug attaching the debdiff.

And I do have a practical question: The current package has the
version number 13.04.2-4+b1. If I upload my new version it will be
13.04.2-4u1. Is this correct?

Close(ish) - 13.04.2-4+deb8u1. (Also upload versions are based on source versions, so the current version in stable is just 13.04.2-4.)
