Others have told you the correct path for this. But there is an alternative
way to change your profile...<

# eselect profile list
Available profile symlink targets:
  [1] default-linux/x86/2006.1
  [2] default-linux/x86/no-nptl
  [3] default-linux/x86/no-nptl/2.4
  [4] default-linux/x86/2006.1/desktop *
  [5] hardened/x86
  [6] hardened/x86/2.6
# eselect profile set 4

Also note that the slash in the end is important:

# ls -l /etc/make.profile/
total 3
-rw-r--r-- 1 root root 655 2006-07-26 22:30 make.defaults
-rw-r--r-- 1 root root 3 2006-07-26 22:30 parent
-rw-r--r-- 1 root root 271 2006-07-26 22:30 virtuals

# ls -l /etc/make.profile
lrwxrwxrwx 1 root root 57 2006-09-09
17:38 /etc/make.profile ->
..//usr/portage/profiles/default-linux/x86/2006.1/desktop
