Yes you can.
(By disconnected, I assume you mean a computer not connected to the 
internet.)

Of course, you will need to ensure that the maps or vectors/images you 
are accessing are on your computer.

Try building a demo which references GML files. (I'm pretty sure 
demo/openlayers/index.html does this still).
