I confirm that this is a bug, which was not there in the version from mid Dec.
It appears that the align key does not have any effect. Here's a simpler
example:
\definemathmatrix[matrix]
[align={right,left,left}]
\starttext
\startformula
\startmatrix
\NC ABC \NC ABC \NC ABC \NR
\NC AAAAAA \NC AAAAAA \NC AAAAAA \NR
\stopmatrix
\stopformula
