What exactly is your setting for org-agenda-files?
Is it a directory, or a one-element list with a directory as the  
element?

a new org item buffer (C-c C-w).

Maybe double-check the value of `org-agenda-file-regexp' so that a
directory in `org-agenda-files' returns something sensible.
