clinical settings? Both Special Stain and IHC? What would you like to see in 
your control slides? What don't you want to see- long lead times, quality? As 
for Research, what are your requirements?

[Histonet] Aspergillus

quality naturally occurring but will accept good quality home grown. We usually

How can you make some of your Histology equipment last forever?

Water bath- fill the water every morning with water at about the same 
temperature that you operate. Every one of these water bath units contain a 
heating element that is the biggest source of failure. The less you make it 
work means you can extend the life of your unit ten times. Heat the water in a 
microwave or on the stove first.

Embedding Center- Same thing. Melt the wax ahead of time in a less draw 
intensive "coffee pot." before you put it in the chamber.  Don't ever crank the 
temperature of the unit up to melt the wax fast so you can get to work quicker. 
The heating elements will fail in three times their life.  Replacing the 
heating element in your embedding center will cost you $600-1000 dollars. 
Replacing the coffee pot $100.

Block Trimmer- same thing. Don't let it run too long when you are not using 
them. They use the exact same silicone heating element. The less it is on the 
longer it lasts.

Cryostat- put it to sleep at minus 10 each night. on weekends turn it off, open 
the top and let it dry out over the weekend. Don't believe the crap about the 
seals drying out if you turn it off. Do they dry out in your car in the Winter 
when the A/C is not running?

[Histonet] Sections sticking to tweezers

Can anyone tell me why my sections are sticking to my tweezers and turning into 
one long string of snot when I try to separate them? I have a good pair of 
curved stainless tweezers and I am trying both the front and the back and 
tapping sharply while allowing the tweezers to open at the separation point. I 
can separate one or two, then it sticks and pulls up 7 or 8 sections into one 
long slimy string. My water bath is set at 38.5 and I am sectioning at 4-5 m. 
Even when I go to 6 it still does it. Any ideas?

I am working with a microtome that has an unusual blade holder.  Standard
low profile blades are too short and are covered by the stage so no cutting
occurs. The standard high profile blades stick out above the blade holder
edge significantly and this causes wobble in the blade while sectioning
(creating thick and thin sections and or skipping sections entirely before
cutting a really thick section).

Can anyone suggest what type of blade I should purchase to use with this
holder?  I was thinking maybe sapphire knife blades might fit but didn't
want to waste money buying items I don't need

[Histonet] Problems with Melan A staining

One of my histotech friends is having issues with Melan A staining. Using 
MA5-27949 Melan A antibody from Thermofisher, the negative Kidney tissue 
control was stained strongly positive while the positive melanoma tissue 
control was stained weakly positive. He couldn’t figure out why after several 
tries to reduce background. Can someone give me some suggestions? Thank you 
very much!

If that were true, all cancer research and antibody development in the USA 
would come to a complete halt. Get a signature from someone who died of cancer 
10 years ago??  Here are the findings of most US courts. Here are the facts.

In legal disputes concerning the research use of excised tissues, courts 
consistently have rejected the idea that patients are the owners of their 
excised tissues or retain any property rights. While only three cases have been 
adjudicated to date, the findings (in both state and federal courts) have been 
consistent: patients do not have individual ownership, retain property rights, 
or have complete control over research use of their excised tissues.9,

[Histonet] Apple Green Birefringence in Amyliod slides

Hi everyone.  I was wondering if anyone out there has any experience with 
diagnosing Amyloid tissue using Congo Red stained Kidney using polarized 
lenses.  Is it common to use polarized light to detect Amyloid deposits?  Does 
the absence of the "apple green birefringence" indicate a problem with the 
control tissue or the control slides?  Should this green bifringence always 
appear to confirm the diagnosis?  I know that the tissue should be cut thicker 
than normal (we usually cut at 5), but in the future maybe we will cut at 7 or 
8?

[Histonet] Gram Staining

We are trying to stain a supposed Gram Positive paraffin Tonsil tissue block 
using Gentian Violet, Iodine (Lugol), Gram's decolorizer, Carbol Fuchsin 
Counter and Tartrazine and are having trouble distinguishing the positive from 
the negative. Anyone have any suggestions?

adhesive process that dries up to 50% faster than a normal Silane coated 
charged slide and we are looking for someone that might be using a Ventana or 
who may be dealing with difficult tissue or a difficult protocol that may be 
causing them to lose tissue who can try them for us in a difficult real world

[Histonet] Question concerning H-Pylori Staining

Hi everyone!  We have a question about staining for H-Pylori Using Quick Stain 
(Periodic acid 1%, Alician Yellow, Sodium Metabisulfate,Toluidine Blue stock, 
Sodium Hydroxide) we notice what clearly  looks like the H-Pylori purple 
stained clusters, but after dehydration in 100% alcohol the purple clusters 
seem to disappear. Should we just dehydrate using a slide oven instead of the 
alcohol? For how long at what temp? Could the alcohol be affecting the purple 
color making it too light to see?

10 years. Many of them can be used by researchers and Pathologists and are 
badly needed. If you don't want to sort them out, we will pay you a fair price

Hello All:

I have thousands of tissue blocks from a large US certified reference lab. Most 
of them were used for IHC diagnosis. I am looking to trade what I have for more 
common Histology blocks for control slides such as Mast Cell, Pneumocystis, 
Copper, Gram, Aspergillus fungus, H-Pylori, Spirochete, Acid Fast Bacteria and 
other common Histology tissue for special stain control slides. I also have 
lots of normal tissue blocks. Please let me know if you are interested.
