Where on the tube is the puncture?  If it's rim-side, check for misaligned rim tape.  If it's tire-side, look for something embedded in the tire.  Nearly invisible wires can get stuck inside a tire, repeatedly causing punctures in the same place.  This is one reason you always align the valve
