Anything is possible if you can figure out a way to do it. I drilled a
hole in the front of a case once to mount a firewire port. In most of
the ATX cases I've seen the front bay brackets are either permanently
riveted, or otherwise permanently attached to the case frame. Of course
there are ways to cut around those. But even if you did figure a way to
mount it inside, wouldn't the front bezel of the case still be only
3.5"? You'd have to cut that larger too.
An alternative would be to buy an external enclosure for about $20 or
$30 and put the tape drive in it. They have EIDE, SCSI, SATA enclosures
that attach to the PC via USB ports.
But if you just wanna keep a running backup of your hard drive, just buy
an external hard drive (way cheaper than the LTO).
