Yes, the unknown cert error indicates you do not have a CA cert for tlsv1.  Did 
you get the VenueServer certificate with the Certificate Manager "Request 
Certificate" button? or did you get it some other way?  The issuer on the 
VenueServer certificate should be:
 /O=Access Grid/OU=agdev-ca.mcs.anl.gov/CN=Access Grid Developers CA

I think you have a cert from ANL that was meant for a different purpose.

I was able to install the certificate in Certificate Manager from ANL whenever 
I already submitted the certificate for Venue Server for a few weeks ago. So, I 
ran the VenueServer3.py in DOS command line and it seems worked through Venue

I'm not sure what the exact problem is.  How did you obtain the certificate 
from ANL?  Were you able to install it?  If so, view the certificate and see 
what the CA designation is, then make sure you have a CA certificate listed in 
the CA certs tab in the Certificate Manager.
