Hi,
If you are trying to do this in a usual mp3 or wmv or other similar file 
formats, it wont be possible as its a single file, wherein the singer's voice 
and the different musical instruments are not given in seperate tracks. For 
this one should have access to the unmix (what they call in the studio), and it 
would have been made in a particular sound editing software such as Nuendo, 
Sound forge etc. This ofcourse would be only with the copyright 
holder/production house of the song.

But you can get the sing-along (karaoke) CDs of some songs in the music stores.

Try the open source audacity editor. It may have the feature you are looking
for.
