I have an AD group and that AD group is assigned permissions in this application Adobe Connect. However when the user tries to access any information it says access denied - When I look at the properties of his account in Connect - he is not showing as a group member But I can confirm that he is a member and this issue appears at first glance to be that the group contains more than 500 users.

Are you referring to the application? I'm somewhat confused by this question. But if I understand your question then remember you can only distribute the application to a user not a computer.
