Check the "DAC/ADC Loopback" switch - that one gave me problems for ages until I spotted it. (Set a sample loop-playing, then start fiddling with alsamixer until you get sound.) Try toggling the "mute" on this item (it's a switch, so this actually turns it on/off) and see if you get sound.
