Yes, this message is within the .EXE file, as part of a stub program that runs
when the program encounters DOS without Windows.  Default stub program displays
a message on screen like "This program cannot be run in DOS mode" or "This
program requires Microsoft Windows", though it could be any DOS program,
including something nefarious.  Probably safer not to execute those things even
in straight DOS.
