Looks like the Tck/Tk wrapper script has not been built. Try running
scons tcltk/asc4dev
and then run
scltk/asc4dev
You can't run 'asc4dev.in' as that's a file into which some key
operating variables have to be inserted (that's what the '.in' indicates.
The other errors are only really warnings arising because you don't have
the CONOPT solver (which is not FOSS, unfortunately). You have a working
version of the PyGTK GUI already by the sound of it.
