I think that if your connection is transformer isolated, the ground
connection will be mostly irrelevant.  However, if not transformer
isolated, the ground can be critical, because the grounds of your 2
systems may literally be may volts away from each other.  Tying the
grounds together will bring the common mode of the 2 systems together,
and allow the amplifiers to work properly.  Without connecting the
grounds, you can easily exceed the common mode voltage specs of the
receiving end of the system, and induce nasty hum.  

The Jensen Transformers web site has a bunch of really good app notes
on hum and other problems with long signal lines.
